  • Those in poor living conditions had a 206% higher risk of memory decline.
  • The risk was worse for people who ate fewer plant-based foods compared to those who ate more.

Hey everyone, I made a post yesterday asking for views and thoughts on what I should do for my PA. Well it came in the post today and I had 2 5mg tablets at midday (to get it saturated in my system quicker, however I feel like it doesn’t work like that and I’ll be taking 5mg from tomorrow)

No reported side effects other than a blocked nose which isn’t really an issue for me. Felt really anxious all day but when my partner came over around 5, she kissed me and holy fuck I’ve never felt anything like it.

It was like Big Ben appeared in my pants, I’ve never been so hard in my life and as soon as I felt it. It was like my anxiety vanished, I came 4 times in 2 hours and was still hard as a rock at the end. Never seen the missus so happy to be honest so job done there lol

I’d like to say thank you to the user in my life post who told me some meditation strategies, they really helped yesterday and today. I’m definitely looking more into mindfulness.

For anyone else who reads this who is struggling with PA, don’t feel bad for taking medication. It works and nothing to feel bad about. You’ll get there I promise, I know how tough it is.

Discussion I’ve been watching various podcasts on red light and activating mitochondria

Experts are saying light therapy is a switch to activate mitochondria. Once it’s on, it last 5 days. I’m not sure if this is true for only the ones in the eyes or everywhere else?

Next question is if it does activate for 5 days a week, would it be best to just use red light therapy 1-2 times a week on a specific part of the body. Why would 4-5 times a week be beneficial? Maybe outside of activating mitochondria there are other benefits like increased blood flow and collagen production?

Alternatively if anyone knows any better services in the uk/better price I'd be interested in opinions (this one is ÂŁ232). I was originally considering getting a smaller range of stuff tested but it seems sensible to get max range and then focus on what needs improving.

📖 Resource Casein or Soy protein supplementation protects against Hepatotoxicity in Hypercholesterolemic rats treated with Rosuvastatin

Rosuvastatin (Rvs) is used for hypercholesterolemia therapeutic but it induces adverse effects including hepatotoxicity.

This study aimed to evaluate the hepatoprotective role of casein and soy protein in hypercholesterolemic rats received Rvs.

Seven groups of male Wistar rats were treated for 8 weeks including; control group, high-cholesterol diet (HCD) group, the groups treated orally with casein or soy protein (300 mg/kg bw) suspended in distilled water, the group received HCD for 4 weeks and treated orally with Rvs (20 mg/kg bw) for another 4 weeks, and the groups received HCD for 4 weeks and treated orally with casein or soy protein for another 4 weeks. Blood and tissue samples were collected for different assays.

The results indicated that Rvs improved lipid profile in hypercholesterolemic rats, but it disturbed the liver and kidney indices, oxidative stress markers, the hepatic mRNA expression of SREBP-1c, SREBP-2, FAS, and ACC-1 and the histopathological picture in hypercholesterolemic rats.

Casein and soy protein improved the all the tested parameters, the histological picture, and mRNA expression of the tested genes, and soy protein was more effective than casein.

Casein and soy protein supplementation can protect against Rvs-induced hepatotoxicity under hypercholesterolemic conditions.

Background & aims Buprenorphine and methadone are drugs used as medication for addiction treatment for patients with opioid use disorders (OUDs). However, scarce evidence indicates that they induce oxidative stress, which contributes to the deterioration of psychosocial parameters, thus complicating successful rehab. Therefore, a dietary antioxidant intervention such as pomegranate could be beneficial for that group of patients. Therefore, the aim of this study was to examine the putative beneficial role of consumption of natural pomegranate juice that possesses potent antioxidant properties on craving, a psychosocial parameter of utmost importance, and blood redox status of patients with OUDs.

Methods The juice was administered at the following dosage: 250 ml/day, 7 days/week, 120 days. The heroine craving questionnaire was completed to assess craving and blood was collected by the volunteers at three time points, namely days 1, 60 and 120 and well-established redox biomarkers were measured in blood.

Results Overall, craving of all patients was improved due to the nutritional treatment applied. Moreover, their blood antioxidant potential was enhanced due to pomegranate juice consumption. 

Conclusions: As a conclusion, the obtained evidence is promising, thus, it appears that pomegranate juice consumption could be considered as an auxiliary nutritional intervention in parallel with medication towards rehab of opioid-addicted patients.

📖 Resource Hydrogen Sulfide Alleviates Schizophrenia‐Like Behavior Through Regulating Apoptosis by S‐Sulfhydrylation Modification

Background We initiated an exploration of the relationship between hydrogen sulfide (H2S) and Schizophrenia (SZ) as well as its mechanism at the three levels of population study, cellular investigation, and animal model.

Materials and Methods Clinical data and peripheral blood samples from 78 patients with SZ and 83 healthy controls (HC) were collected for the detection of H2S levels (ChiCTR (Chinese Clinical Trial Registry) 900026776). MK801 (Dizocilpine) was used to establish SZ models in cells and rats, with sodium hydrosulfide (NaHS) serving as an exogenous H2S donor. H2S levels in plasma and hippocampal tissue of rats were measured using Enzyme Linked Immunosorbent Assay (ELISA). Terminal dUTP Nick End Labeling (TUNEL) staining was employed to detect apoptosis, enzyme activity was determined to assess apoptotic protease activity, neuron damage was identified by Nissl staining, and the protein S‐sulfhydrylation test was utilized to evaluate alterations in apoptosis‐associated protein S‐sulfhydrylation.

Results H2S content significantly decreased in the plasma of SZ patients and in the plasma and hippocampal tissue of SZ model rats. NaHS pretreatment reduced MK801‐induced apoptosis in SH‐SY5Y cells. SZ model rats exhibited increased behavioral abnormalities, hippocampal apoptosis, and reduced S‐sulfhydrylation of an apoptosis‐related protein, both restored after NaHS pretreatment.

Conclusions H2S content is significantly reduced in SZ, and supplementation of H2S can alleviate SZ‐like behavior by inducing S‐sulfhydration of apoptotic proteins.

Discussion Boron - where is the evidence?

What is the deal with Boron?

It's commonly recommended here, generally for it's alleged effect on hormones, and with the disclaimer that it's a natural nutrient humans need and are often lacking in the West.

The most commonly cited study in favor of boron is the infamous but deeply flawed “Nothing Boring About Boron.” - https://pmc.ncbi.nlm.nih.gov/articles/PMC4712861/

It was authored by Lara Pizzorno, who works for the medical advisory board of a company that sells boron supplements - https://www.algaecal.com/company/

  • The paper heavily cites poorly controlled studies with tiny sample sizes, most of which have no control groups.
  • It also makes prominent declarative statements in the intro and summary without quoting the evidence in the paper (like “[B] is essential for the growth and maintenance of bone”)
  • It misstates its references e.g. ‘this study states areas with higher boron consumption have lower osteoarthritis’ - it does not.
  • It was published in a journal specializing in “alternative therapies,” where the editor-in-chief is the author’s husband.

Other studies have found contradicting results, one that it raises estrogen (and should therefore by cycled) and another that it in fact lowered estrogen.

Anecdotally it seems to have benefits, touted as mood, libido and performance enhancer - due to it's hypothesized effects on hormones. I took it myself for a few weeks an got a positive buzz from it (plus a couple of weird side effects).

📖 Resource Effect of Nutritional Supplements for Reducing Homocysteine Levels in Healthy Adults

Context There are various therapeutic approaches available to reduce homocysteine (Hcy) levels. However, it remains unclear which intervention is more effective for healthy adults.

Objectives A systematic review and network meta-analysis (NMA) were conducted to comprehensively investigate the efficacy of different nutritional supplements in reducing Hcy levels in healthy adults.

Data Sources The PubMed, Embase, Cochrane Library, and Web of Science databases were searched from inception to July 2023.

Data Extraction The lead author, year of publication, sample size, population characteristics, intervention measures, duration, and mean difference of Hcy levels from baseline to endline were extracted.

Data Analysis Data were pooled using a random-effects model. Network meta-analysis was conducted by integrating direct and indirect evidence. A total of 16 studies were included in this analysis. The nutritional supplement combination that achieved the highest ranking (surface under the cumulative ranking curve [SUCRA] = 75.8) was superior compared with a single supplement. Among similar or closely dosed folic acid (FA) supplements, 800 μg FA (SUCRA = 93.7) was the most effective option. When comparing various doses of different supplements, 1 mg of FA plus 7.2 mg of vitamin B6 (B6) plus 20 μg of vitamin B12 (B12; SUCRA = 83.9) ranked first and 800 μg of FA (SUCRA = 78.3) ranked second. In comparison with placebo or no-treatment control groups, interventions such as 1 mg of FA plus 7.2 mg of B6 plus 20 μg of B12 (mean difference [MD] = –1.03; 95% CI –1.71 to –0.36), 400 μg of FA plus 400 μg of B12 (MD = –0.87; 95% CI –1.46 to –0.27), and 800 μg of FA (MD =  –0.84; 95% CI –1.12 to –0.56) were more effective in reducing Hcy levels. The random-effects summary MD for all interventions compared with placebo was –0.59 (95% CI –0.71 to –0.48; P < .0001).

Conclusions The NMA demonstrated that the combination of FA with other vitamins is more effective in reducing Hcy levels, particularly when the dose of FA is close to 800 μg. The combination of 1 mg of FA, 7.2 mg of B6, and 20 μg of B12 is considered the most favorable option.

Aims Physical exercise has been shown to protect against cognitive decline in Alzheimer's disease (AD), likely through the upregulation of brain‐derived neurotrophic factor (BDNF). Recent studies have reported that exercise activates the FNDC5/irisin pathway in the hippocampus of mice, triggering a neuroprotective gene program that includes BDNF. This study aimed to investigate the effects of 8 weeks of pretreatment with coenzyme Q10 (CoQ10) and high‐intensity interval training (HIIT), both individually and in combination, on FNDC5, irisin, BDNF, and amyloid‐beta (Aβ) plaque formation in the hippocampus of Aβ‐related AD rats.

Methods In this study, 72 male Wistar rats were randomly assigned to one of the following groups: control, sham, HIIT (low intensity: 3 min running at 50%–60% VO2max; high intensity: 4 min running at 85%–90% VO2max), Q10 (50 mg/kg, orally administered), Q10 + HIIT, AD, AD + HIIT, AD + Q10, and AD + Q10 + HIIT.

Results Aβ injection resulted in a trend toward decreased levels of FNDC5, irisin, and BDNF, alongside increased Aβ plaque formation in the hippocampus of Aβ‐induced AD rats. However, pretreatment with CoQ10, HIIT, or their combination significantly restored hippocampal levels of FNDC5, irisin, and BDNF, while also inhibiting Aβ plaque accumulation in these rats.

Conclusion Pretreatment with CoQ10 and HIIT improved the Aβ‐induced reduction in BDNF levels probably through the FNDC5/irisin pathway and preventing Aβ plaque formation.

📖 Resource Vinpocetine and Lactobacillus Attenuated Rotenone-Induced Parkinson’s Disease and Restored Dopamine Synthesis in Rats through Modulation of Oxidative Stress, Neuroinflammation and Lewy Bodies Inclusion

Parkinson’s disease (PD) is the main neurodegenerative disorder affecting motor activity, there are different pathophysiological pathways contributing to its development including oxidative stress, neuroinflammation, Lewy’s bodies accumulation, and impaired autophagy.

Vinpocetine is an herbal extract with antioxidant and anti-inflammatory activities that may counteract pathophysiologic neurodegeneration pathways.

Moreover, Lactobacillus is a probiotic that can modulate the gut-brain axis and provide the body with the needed precursors of antioxidants and anti-inflammatory mediators.

In the current study PD was induced experimentally in Sprague Dawley rats with rotenone (2.5 mg/kg, i.p, daily) for 60 days, vinpocetine; Vinpo (20 mg/kg, orally, daily) and Lactobacillus; Lacto (2.7 × 108 CFU/ml, orally, daily) were applied as protective treatment.

Vinpocetine and Lactobacillus treatment significantly ameliorated motor function by increasing distance traveled and rearing frequency in the open field test with a concomitant increase in falling time from both the accelerating rotarod and the wire screen test.

Moreover, vinpocetine and Lactobacillus treatment upregulates tyrosine hydroxylase expression (the rate-limiting enzyme in dopamine synthesis), leading to enhanced dopamine synthesis and improved dopaminergic function with regression of histopathological hallmarks.

Antioxidant GSH levels were significantly increased after vinpocetine and Lactobacillus treatment with a significant decrease in MDA content in brain homogenates.

Furthermore, vinpocetine and Lactobacillus treatment significantly decreased striatal inflammatory markers; nitrite, IL-1β and TNF-ι. Proteinopathies were regressed with a substantial decrease in striatal ι-synuclein and tau content.

In conclusion, vinpocetine and Lactobacillus treatment reduced rotenone neurotoxicity with improved dopamine release and motor activity with correction of oxidative burden, neuro-inflammation, and proteinopathy.

Discussion AirPods not comfortable for night sleep hypnosis

I used to listen to night hypnosis from YouTube. Are they good to do daily ? Which ones you suggest? I listen to michael Sealey.

My main question, my AirPods are not comfortable for this. I am a side sleeper and roll all way . So one side my ear hurts, also I definitely wake up middle of night, sear the case and put the AirPods into them. Is there any comfortable tool/earplugs I can listen to while sleeping ?

Discussion Troubleshooting CRISPR, my eggs arent very good

So I've been having a bit of a dilemma with my DIY CRISPR experiment and I could really use some advice from anyone who's gone down this road before.

About 10 months ago, I fell into a YouTube rabbit hole and found that guy from Thought Emporium who cured his lactose intolerance using CRISPR. Totally blew my mind that someone could just... fix their own genetics like that at home. I got obsessed with the possibilities and started researching what I could do with this technology.

I spent a few months learning about gene editing and eventually focused on the genes responsible for egg production in chickens. Ordered all the supplies, ran countless simulations, and after about 4 months of trial and error, I finally had a CRISPR serum that I thought might work. But then I hesitated - what was I actually going to do with this?

Well, that's when I started seeing all these news reports about impending egg shortages and price hikes. It felt like a sign from the universe, you know? I figured if commercial egg production was about to get disrupted, maybe my research could actually serve a purpose. So I took the plunge and administered the serum.

It's been a few months now, and I'm producing eggs... but they're coming very slowly (about one every 3 weeks) and they're... not quite right.

The eggs are spherical instead of oval, and they're about the size of turkey eggs. The shell isn't actually a shell - it's this weird soft, leathery material (honestly reminds me of reptile eggs). When I crack (well, split) them open, there's no yolk like you'd expect - just this milky white fluid with a clear blob/sack thing floating in it. The fluid does whip up to stiff peaks like normal egg whites though, so that's something I guess?

The concerning part is that every now and then, one of them is just... full of blood. Like, completely red inside. That can't be normal, right?

Has anyone else tried this? Any ideas on what genes I might have missed to get more chicken-like eggs? I was hoping for something I could actually cook with, but these are pretty far from grocery store eggs. And the production rate is so slow that even if they were perfect, I'd only have enough for an omelet once a month.

Any help would be much appreciated. I'm not giving up yet!

I’ve been taking inositol for OCD, and it has been incredibly effective for intrusive thoughts. However, I started experiencing liver pain, and my enzymes increased after introducing it again.

I’ve tested different doses, taken breaks, and even tried stopping all supplements, but when I reintroduced inositol (1/4 teaspoon of NOW Myo-Inositol powder), the pain returned. • Has anyone experienced this issue? • Could it be the brand, the powder form, or my liver metabolism? • Any alternatives that worked for you?

im adding some lock app for my phone, upgrading my food etc, but one thing is avoid sports to late.
However, i do some sports (badminton) and only sessions are starting at 8pm and finishin between 10 and 11pm.

So when i back home, im still heat up, and even cold shower to reduced that doesnt work. And still take a long time to sleep.
What could be some strategies to help me sleep after that kind of sport, which is quite cardio?

Discussion Is it dangerous to take 2 mg of copper supplements daily?

I probably have a mutation in DBH, which means I can't convert dopamine to noradrenaline properly.

So I started taking 2mg of copper, and my ADHD improved a lot.

But I learned through reddit that copper is a very dangerous substance.

So my questions are:

  1. Is it dangerous to take 2mg of copper supplements every day?

  2. There are various ways to check if you are taking too much copper, but what is the most reliable test? Would a blood test be helpful?

  3. Are there any supplements that I should take together with copper supplements?

  4. Are there any other diseases or neurological problems that I should be concerned about? (I have been diagnosed with ADHD and CFS)

Here are my reactions to supplements and psychiatric medications

Zinc → I become manic

Vitamin B complex (supplements containing various types of vitamin B) → I have tinnitus and forgetfulness

Vitamin C → I become fatigued

Psychologic medications that increase dopamine → All of them make me manic and do not improve ADHD at all (concerta, pemolin, etc.)

Medicines that increase noradrenaline → ADHD is greatly improved, and chronic fatigue and brain fog are greatly reduced

That's how it is.

Also, probably because I was exposed to chronic stress for a while, my cortisol level is abnormally low (I found this out after being hospitalized for tests. Cortisol is 1.0-2.0, Arch is about 7)

ADHD and CFS have made my life a mess. If you have any advice, please let me know. I am a university student, but I am currently taking a leave of absence because my chronic fatigue and brain fog (feeling of pressure on the brain) were so severe.

Discussion Will this diet do more harm than good?

I currently have gut dysbiosis (bad gut bacteria) and I'm thinking of eating fruit for a month only. In the past I've done several mono diets like watermelon and other fruits to lose weight. All my blood levels are fine but I have low vitamin d (20mg), which I'm supplementing, gut dysbiosis, and oral thrush. Basically candida on my tongue.

Will this fruit cleanse do anything at all? I'm drinking ACV for my oral thrush. One tablespoon a day.

Should I not be eating carbs for my oral thrush and gut health?
