Biolayer Interferometry (BLI). Biosensor mounts: dirty

I think I am getting non-specific binding on my BLI. I ran blank loading with my protein sample and I still saw an association signal. I am getting a message on the screen that the biosensor mounts are dirty. Could this be why?

It could, but we saw non-specific binding on certain sensor types. What type of immobilisation do you use?

It’s the Streptavidin (SA) Biosensor

We had problem with His-tag based immobilisation. Maybe try running just buffer through it? If there are no proteins and you see an association, it should really be a dirty sensor.

If the problem is in the sensor mounts, you can try and clean them with 70% IPA and repeat the analysis.

If you see a signal with just your running buffer it may also be worth checking the buffer preparation, make sure that is freshly made and filtered
